For fluorescence application, the exposure time is long and the frame rate is very slow. It take a long time and is very difficult for users to find the focus. Sometime it is not acceptable because some kinds of fluorescence quenching are very fast. There are some methods to help to resolve this problem:
Lower the exposure time to increase the frame rate. At the same time, adjust the gain settings to increase the brightness.
If the above methods is regarded as too noisy or still too dark to see the object, digital binning in additive mode will help to increase the brightness with the sacrifice of resolution. 2*2 will increase the brightness to 4 times of the original one and 4*4 digital binning will increase the brightness to 16 times.
After find the focus and when users want full resolution snapshot with certain gain setting, users could estimate the exposure time according to the current exposure time, gain and digital binning settings.
